Recent advances in brain imaging and spectroscopy techniques allow scientists to examine brain activity and its underlying biophysical basis in vivo, during normal and altered physiological conditions. As a result, scientists can now utilize these methods to better understand the neurophysiological basis of drug addiction. Here we review how several cutting-edge approaches have been used to examine the brain networks involved in drug-seeking and drug-taking behaviour and to probe the relationships among neural activity, drug use and drug addiction. We begin with methods that allow researchers to probe brain function during natural and drug-induced states of consciousness. We then review applications of new magnetic resonance imaging techniques to investigate the neuroanatomical correlates of addiction-related behaviours and to monitor addiction-related changes in brain structure. Finally, we consider advances in spectroscopy and in vivo magnetic resonance spectroscopy to study how drugs alter specific neurochemical processes in vivo. Taken together, the methods reviewed here provide new insights into the neural substrates of addiction and should enable a better understanding of how drugs of abuse modulate neural circuits and mechanisms involved in addiction.Genetic counseling and testing for Huntington's disease in an American Indian population.
